HyClone CultiSpher S Microcarriers

Thermo Scientific* HyClone CultiSphers Microcarriers are unique protein-based macroporous microcarriers, manufactured from pharmaceutical grade porcine gelatin via a process.
CultiSphers may also be utilized as an excipient component in preparations of cellular products intended for implantation. Use of the microcarrier in these cellular therapeutic products has been shown to enhance the survival of the implanted cells. CultiSpher microcarriers are made of gelatin. Gelatin is a product derived from the partial hydrolysis of collagen and have large regions of identical structure. The majority of primary cells will attach to the gelatin structure. Since the macroporous microcarrier structure creates a three-dimensional framework it mimics the cooperative conformation demonstrated in tissue. The macroporous structure and the digestible gelatin matrix allows the cells to increase from 10 to 20 to 2000 to 3000 on each bead. As the matrix is easily digested with tissue culture grade trypsin all these cells can be recovered and used for scale-up purposes.

When used in cell cultures, cells can attach to both the external and the internal surfaces of the matrix. The increased surface area of the matrix together with the protection from stress afforded to the cells in the interior of the matrix results in enhanced cell production capabilities. An additional advantage of the product is that the matrix can be dissolved with proteolytic enzymes resulting in in the harvesting of cells with almost 100% viability.

  • Cavernous structure allows for significantly greater numbers of cells per bead (> 2000 cells per bead)
  • Highly cross-linked matrix and interior surfaces better protect cells from shear stress
  • Because gelatin is derived from collagen, cells attach more readily and proliferate in an environment that more closely approximates an in vivo environment
  • Complete dissolving of gelatin matrix with trypsin yields a 95% to 100% recovery
  • Easily scaled-up by 50-fold increments from 1 to 50, for up to 2500L bioreactors
