The hydrolysis of the non-fluorescent Cellomics™ Live Cell Stain Blue by intracellular esterases generates the strongly fluorescent hydrophilic Cellomics Live Cell Stain Blue that is well retained in the cell cytoplasm. The esterase activity is proportional to the number of viable cells, and the fluorescence intensity generated from the esterase-catalyzed hydrolysis of the stains can also be used to measure the cell number.
